ChIP-exo protocol. The protocol utilizes adapter ligation and lambda exonuclease while the DNA is still crosslinked to the protein. After elution of the DNA and creation of double-stranded DNA (dsDNA), a unique adapter is ligated to the other end, thereby marking the 5' ends of the DNA-protein interactions. dsDNA, double-stranded DNA; TF, transcription factor.
